Activator protein 1 (AP-1) was first discovered as a 12-O-Tetradecanoylphorbol-13-acetate (TPA)-activated transcription factor that bound to a cis-regulatory element of the human metallothionein IIa (hMTIIa) promoter and SV40.[1]

Consensus sequences

The AP-1 binding site was identified as the TPA response element (TRE) with the consensus sequence 5’-TGA G/C TCA-3’.[2]

12-O-tetradecanoylphorbol-13-acetate (TPA) responsive element (TRE) (TGAGTCA).[3]

Samplings

Copying the consensus sequence for the DNA-binding factor (TRE): 3'-TGAGTCA-5' or 3'-TGACTCA-5' and putting the sequence in "⌘F" does not find either sequence in either A1BG promoter using nucleotide positions as can be found by the computer programs.

For the Basic programs testing consensus sequence 3'-TGA(G/C)TCA-5' (starting with SuccessablesTRE.bas) written to compare nucleotide sequences with the sequences on either the template strand (-), or coding strand (+), of the DNA, in the negative direction (-), or the positive direction (+), the programs are, are looking for, and found:

  1. negative strand in the negative direction (from ZSCAN22 to A1BG) is SuccessablesTRE--.bas, looking for 3'-TGA(G/C)TCA-5', 0.
  2. negative strand in the positive direction (from ZNF497 to A1BG) is SuccessablesTRE-+.bas, looking for 3'-TGA(G/C)TCA-5', 0.
  3. positive strand in the negative direction (from ZSCAN22 to A1BG) is SuccessablesTRE+-.bas, looking for 3'-TGA(G/C)TCA-5', 0.
